ABSTRACT

Objective:To investigate the effect of milk on sebum secretion in golden hamsters, and to explore its possible mechanism of action. Methods:Eighteen golden hamsters were randomly and equally divided into 3 groups: blank control group receiving no intervention, whole-milk group gavaged with whole milk, and skimmed-milk group gavaged with skimmed milk. The gavage feeding was performed at a dose of 2.5 ml twice a day for 4 consecutive weeks. The maximum transverse diameter and maximum longitudinal diameter of bilateral sebaceous gland spots were measured on days 0, 7, 14, 21 and 28 after the start of intervention, and the area of sebaceous gland spots was calculated; at 24 hours after the last gavage, bilateral sebaceous gland spot tissues were resected, and subjected to immunohistochemical study to determine the expression of insulin-like growth factor-1 (IGF-1) /sterol regulatory element-binding protein-1 (SREBP-1) /acetyl-coenzyme A carboxylase (ACC-1) signaling pathway in sebaceous gland spots. Statistical analysis was carried out by using repeated measures analysis of variance, one-way analysis of variance for independent groups, Kruskal-Wallis H test, and least significant difference- t test for multiple comparisons. Results:Repeated measures analysis of variance showed that there was no significant difference in the area of sebaceous gland spots of golden hamsters among the 3 groups ( F= 0.96, P= 0.417) . The IGF-1 expression was significantly higher in the skimmed-milk group (0.39 ± 0.03) than in the blank control group (0.35 ± 0.03, t= 2.62, P= 0.021) and whole-milk group (0.33 ± 0.02, t= 3.82, P= 0.002) ; compared with the blank control group (0.36 ± 0.02) , the skimmed-milk group showed significantly increased SREBP-1 expression (0.42 ± 0.04, t= 2.64, P= 0.021) ; the ACC-1 expression was significantly higher in the skimmed-milk group (0.40 ± 0.03) and whole-milk group (0.40 ± 0.05) than in the blank control group (0.34 ± 0.03; t= 2.39, 2.47, P= 0.031, 0.026, respectively) . Conclusion:Milk may promote sebum secretion in golden hamsters through the IGF-1/SREBP-1/ACC-1 signaling pathway.

ABSTRACT

Objective:To evaluate the effect of pomegranate peel polyphenols on sebum secretion in golden hamsters, and to explore its possible mechanisms. Methods:Thirty golden hamsters were randomly and equally divided into 5 groups: (ointment) vehicle group, 0.48%-, 0.96%-, 1.92%-pomegranate peel polyphenol ointment groups, and retinoic acid cream group. Corresponding cream or ointments were applied to bilateral sebaceous gland spots of the golden hamsters at a dose of 1 gram twice a day for 4 consecutive weeks. The area of bilateral sebaceous gland spots was measured on days 0, 7, 14, 21 and 28 after the start of treatment, which was calculated by the maximum longitudinal diameter multiplied by the maximum transverse diameter. Twenty-four hours after the last treatment, immunohistochemical study was conducted to determine the expression of AKT/Sox9 signaling pathway in sebaceous gland spots resected from the golden hamsters. The area of sebaceous gland spots in these groups at different time points was compared by repeated measures analysis of variance, and other data were analyzed by one-way analysis of variance or Kruskal-Wallis H test. Results:The area of sebaceous gland spots was significantly smaller in the 0.96%-, 1.92%-pomegranate peel polyphenol ointment groups (50.48±2.41 mm 2, 48.24±2.56 mm 2, respectively) and retinoic acid cream group (48.31±2.76 mm 2) than in the vehicle group (57.99±3.29 mm 2; t=2.69, 3.98, 3.65, P=0.012, 0.001, 0.001, respectively) . Sox9 expression was significantly lower in the 1.92%-pomegranate peel polyphenol ointment group (0.39±0.04) and retinoic acid cream group (0.38±0.03) than in the vehicle group (0.44±0.02, P=0.040) . However, there was no significant difference in AKT expression among the 5 groups ( F=1.645, P=0.199) . Conclusion:Pomegranate peel polyphenols can reduce the sebaceous gland spot area and inhibit sebum secretion in golden hamsters, which may be related to the inhibition of Sox9 expression.

ABSTRACT

BACKGROUND: Although sebum secretion is crucial for acne development, acne lesion distribution is not always similar to the topographic differences of sebum secretion. OBJECTIVE: To analyze whether sebum secretion affects acne development and distribution and to assess other factors possibly influencing the relationship between acne and sebum secretion. METHODS: This single-center retrospective study included 67 acne patients and 50 controls. Acne patients were divided into 3 groups based on acne lesion distribution: T-zone dominant, U-zone dominant, and mixed groups. The secreted sebum level in each zone of acne patients was compared with that of controls. We also conducted correlation analysis between secreted sebum level and acne number, depending on the facial zone. RESULTS: No significant difference was found between acne patients and controls regarding age and sex ratio. The U-zone dominant group showed increased sebum levels compared with controls in the U-zone and whole face, but a similar result was not obtained in the T-zone dominant group. Moreover, there was a significant correlation between the number of lesions and secreted sebum level in the U-zone, but not in the T-zone. Further, there was a more significant relation in the U-zone of male and young patients. CONCLUSION: We found that increased sebum secretion compared with the condition may affect acne development, especially in the U-zone. Sex and age may also influence the relationship between acne and increased sebum secretion. Acne lesion distribution may vary from patient to patient because sebum secretion affects acne differently depending on multiple factors.

ABSTRACT

@#Parkinson’s disease is a neurodegenerative disease characterized by motor and nonmotor symptoms. Skin manifestations may be seen in patients due to autonomic dysfunction or iatrogenic reasons. In this study, we aimed to identify various skin disorders and compare the skin sebum and moisture percent ages in patients with Parkinson’s disease with healthy controls. Methods: Forty patients with Parkinson’s disease and 40 healthy controls were included in the study. Dermatological history of the patients and dermatological examination were recorded. A portable pen-shaped LCD Display Digital Skin Moist Oil Analyzer (Reyoung-Beauty, Guangdong, China) was used to measure the sebum and the moisture in the skin. Results: In this study, the skin disorders consisted of acneiform and sebum related changes, seborrheic dermatitis, anterolateral leg alopecia, and angular cheilitis, bullous pemphigoid and melanoma. Increased sweating and seborrhea were frequent complaints. While skin sebum content was significantly higher in patients compared to the control group, skin moisture did not differ significantly between the groups. Cutaneous malignancy or iatrogenic skin disorders were not detected in any patient.

ABSTRACT

Background: Biophysical parameters of skin such as trans-epidermal water loss (TEWL), hydration, elasticity, pH, and sebum reflects it functional integrity. Advances in technology have made it possible to measure these parameters by non-invasive methods. These parameters are useful for the prediction of disease and its prognosis. It also helps in developing new skin care products according to various skin types, and to evaluate, modify, or compare the effects of existing products. Aim: The aim of the study was to measure, evaluate, and analyze variations in biophysical parameters at pre-selected skin sites in healthy Indian volunteers, across different age groups and gender. Methods: The study was conducted among 500 healthy Indian volunteers, between 5 and 70 years of age, in the outpatient department of dermatology at Sir T. Hospital, Bhavnagar. Biophysical parameters such as TEWL, hydration, elasticity, and sebum content was measured on four pre-selected body sites by a Dermalab instrument (Cortex Technology, Denmark). The skin pH was measured with a sensitive pH probe (BEPL 2100). Results: All parameters were higher in males compared to females, except for sebum content, which was equal in both genders. Transepidermal water loss and hydration was lower in middle and older age groups. The skin pH showed no statistically significant difference with age. Sebum content was higher in middle and older age groups. The nose had the highest sebum content across all age groups. The forehead showed higher median values of TEWL and hydration compared to other sites. Though elasticity has highest value on forearm, only leg region showed statistically significant value. Limitations: The present study was confined to a single geographical area, so the effect of environment changes could not be judged accurately. Seasonal variations were not studied as it was a cross-sectional study. Conclusion: Skin properties vary with age, gender, and location on the body. This knowledge will help to create a database of these parameters in the Indian population. It would assist in the diagnosis of various clinical conditions and monitor therapeutic response.

ABSTRACT

ABSTRACT Green tea (Camellia sinensis (L.) Kuntze, Theaceae) polyphenols have activities against skin disorders; however, anti-sebum efficacy of green tea cosmetic has sparely been reported. The facial tonner containing green tea was therefore developed and clinical evaluated. The base formulas which stabled following accelerated tests were sensorial assessed in ten volunteers. The base with hydroxyethyl cellulose, glycerin and panthenol (totally 3.6%) with the significant (p < 0.05) preference (82.3 ± 0.55%) over the others was further developed to green tea preparations. All of the products were stable and caused none of skin irritation as closed patch tested in twenty volunteers. Thereafter, they were clinical evaluated in the same group of the volunteers and monitored with Sebumeter® by means of a split-face, randomized single-blind, placebo-controlled study. Anti-greasy efficacy of 2, 4.5 and 7% green tea tonners were 3.47 ± 0.10, 8.18 ± 0.44 and 17.87 ± 0.46% following 14 days of facial treatment. The efficiency was pronounced at the end of the study; day 28 (8.48 ± 0.13, 20.26 ± 1.03 and 31.57 ± 1.22%). Anti-sebum efficacy of the 4.5 and 7% green tea tonners were significantly better than the base formula (day 14; p < 0.05, day 28; p < 0.01). The efficacy of 28 days treatment was significantly better than 14 days (p < 0.05). The safe and efficient green tea tonner for oily face treatment was therefore approved in this context.

ABSTRACT

BACKGROUND: Although many therapeutic agents have been developed, only a few drugs are known to target multiple pathogenic factors in the treatment of acne. OBJECTIVE: The purpose of this study was to identify a new drug candidate, platycodin D, which is a substance extracted from the root of Platycodon grandiflorum. METHODS: Using western blotting and Cell Counting Kit-8 assay, we studied the effects of platycodin D on SEB-1 sebocytes, fibroblasts, and keratinocytes. We investigated its effects in view of lipogenesis, collagen production, anti-inflammatory activity, and dyskeratinization. RESULTS: In SEB-1 sebocytes, platycodin D showed a sebosuppressive effect by downregulating ERK and insulin- like growth factor-1R/PI3K/Akt/sterol-regulatory element binding protein-1 signaling pathways. In addition, adiponectin, one of the adipokines responsible for sebum production, was decreased in platycodin D-treated SEB-1 sebocytes. In fibroblasts, platycodin D increased collagen production and reduced inflammation by inhibiting nuclear factor kappa B and matrix metalloproteinases. Platycodin D also showed anti-inflammatory effects on keratinocytes. It also suppressed keratin 16 expression induced by lipopolysaccharide. Furthermore, platycodin D showed no cytotoxicity on both SEB-1 sebocytes and fibroblasts. CONCLUSION: Our data demonstrate the clinical feasibility of platycodin D for acne treatment and the prevention of acne scarring by sebosuppressive and anti-inflammatory effects, as well as through an increase in collagen levels.

ABSTRACT

Dyssebacia is the name coined to describe numerous plugs of inspissated sebum projecting from dilated orifices of sebaceous glands. This is a case series report of 12 patients who presented to our department with complaints of asymptomatic skin lesions which started first in lower part of nose followed by involvement of entire nose, cheeks and forehead .On examination there were multiple yellow plugs of sebum projecting out from follicular orifices resembling those of shark skin. Dermoscopic examination and Urinary levels of Niacinamide levels in urine were estimated in few cases. Based on history, clinical examination and laboratory, Diagnosis of dyssebacia was made and patients were started on niacinamide following which there was improvement in lesions in 8 of the 12 patients within two to four weeks. Diagnosis of pellagra is based on clinical criteria. Dyssebacia can be the early cutaneous marker of niacin deficiency. Thus in this context detection of dyssebecia acts as a primeval marker in diagnosis of pellagra, which still exists in epidemic proportions in many countries.

ABSTRACT

BACKGROUND: Acne vulgaris has been linked to the Western diet. Hyperglycemic diet increases insulin and insulin-like growth factor (IGF)-1. Deeper insights into IGF-1-mediated signal pathway are critical importance to understand the impact of Western diet. OBJECTIVE: We investigated the effect of IGF-1 on the expression of inflammatory biomarkers and sebum production in cultured sebocytes. METHODS: Polymerase chain reaction and enzyme-linked immunosorbent assay were performed to measure changes in the expression of inflammatory biomarkers including interleukin (IL)-1β, IL-6, IL-8, tumor necrosis factor (TNF)-α, nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), IGF1R, IGFBP2, sterol response element-binding protein (SREBP), and phosphatidylinositol-4,5-bisphosphate 3-kinase, catalytic subunit alpha (PI3KCA) after the treatment of cultured sebocytes with 10⁻⁷ M or 10⁻⁵ M IGF-1. Sebum production was evaluated after the treatment of cultured sebocytes with 10⁻⁷ M or 10⁻⁵ M IGF-1 using lipid analysis. RESULTS: The expression levels of IL-1β, IL-6, IL-8, and TNF-α in cultured sebocytes after treatment with 10⁻⁷ M or 10⁻⁵ M IGF-1 were increased. Increased gene expression levels of NF-κB in cultured sebocytes were also shown after 10⁻⁷ M or 10⁻⁵ M IGF-1 treatments. Gene expression of these inflammatory biomarkers was decreased after 10⁻⁷ M or 10⁻⁵ M IGF-1 treatment in the presence of 100 nM NF-κB inhibitor. Treatment with 10⁻⁷ M or 10⁻⁵ M IGF-1 increased the gene expression levels of IGF1R, IGFBP2, SREBP and PI3KCA in cultured sebocytes. Sebum production from cultured sebocytes treated with 10⁻⁷ M or 10⁻⁵ M IGF-1 was also increased. CONCLUSION: It is suggestive that IGF-1 might be involved in the pathogenesis of acne by increasing both expression of inflammatory biomarkers and also sebum production in sebocytes.

ABSTRACT

BACKGROUND: The clinical symptoms of facial and truncal acne differ. Skin surface acidity (pH), which is affected by sebum secretions, reflects the different clinical characteristics of the face and trunk. However, no studies have been conducted on truncal sebum production and skin pH. OBJECTIVE: We evaluated the differences and relationship between pH values of the face and trunk. We also evaluated the relationship between pH and the quantity of sebum produced in the trunk. METHODS: A total of 35 female patients clinically diagnosed with truncal acne were included. We measured pH on the face and truncal area using the Skin-pH-Meter PH 905®. We measured truncal sebum secretions using the Sebumeter SM 815®. Statistical analysis was performed to evaluate the correlations and differences between pH and sebum. RESULTS: Facial pH was significantly higher than chest and back pH values. The correlation between pH on the trunk and the face was significant. We used linear regression equations to estimate truncal pH using only measured pH from the chin. There was no significant relationship between truncal sebum secretion and pH. CONCLUSION: This was the first study that evaluated the differences and correlations between facial and truncal pH. We found that facial pH can predict truncal pH. In addition, we conclude that differences in pH and sebum secretion between the face and trunk are one of the reasons for differences in acne symptom at those sites.

ABSTRACT

Objective To assess the biophysical characteristics of normal skin and inflammatory skin lesions in mild and moderate acne. Methods Seventy-five mild and moderate acne vulgaris patients were included in the study. One inflammatory lesion measuring 2.0 to 5.0 mm in diameter which occurred within 48 hours prior to the measurement was selected as the target lesion. Trans-epidermis water loss (TEWL), capacitance and a* value were measured in target lesions and lesion-adjacent normal skin. Sebum content immediately,1 and 4 (saturated) hours after face washing was also determined on target lesions and central forehead between the eyebrows. Sebum secretion rate (SSR) was calculated. Results The TEWL and SSR significantly increased in the lesion-adjacent normal skin with the increment of inflammatory lesion number (both P < 0.05), and increased in target lesions with the elevation of a* value (both P < 0.05). The target lesions exhibited a significantly higher TEWL but a lower saturated sebum content and SSR than the adjacent normal skin did (all P <0.05). Conclusions The severity of inflammatory lesions is correlated with the impaired barrier function and increased SSR in facial skin. Compared with the adjacent normal skin, inflammatory skin lesions have a reduced skin barrier function and SSR.

ABSTRACT

This study was performed to analyze the relationship among sebum . hydration content of the skin and nutritional intake, serum antioxidant minerals and antioxidant enzymes, and lipid peroxide concentration in 50 female subjects in their 20s. The skin type was divided into Dry Skin, Mixed Skin, and Oily Skin, and the dry skin group was 14%, the mixed skin group was 56%, and the oily skin group was 30% of all subjects. The average age of the subjects was 20.54 +/- 1.43 years and BMI was 20.66. The average sebum content in each group was in the order of T-zone>forehead>chin>cheek. In case of the T-zone, a significant difference between the dry skin group and the oily skin group was observed, suggesting that the area is most sensitive to sebum content by skin type. Significant differences were not observed in energy and nutrient intakes by skin type. Serum concentrations of antioxidant minerals such as copper, manganese, zinc and selenium were not significantly different among the groups, but the dry skin group tended to be higher than the oily skin group. Serum catalase was significantly higher in the oily skin group (P < 0.05), and MDA was significantly higher in the mixed skin group (P < 0.05). The hydration of the cheek and serum zinc showed a negative correlation, and the sebum content of the cheek and GPx showed a significant negative correlation. The hydration of the forehead and serum copper showed a significant negative correlation, and the hydration of the forehead and GPx showed a significant positive correlation. The hydration of the chin and serum SOD showed a significant positive correlation. With these results, it is considered that the basic condition of nutritional status can affect the skin health.

ABSTRACT

BACKGROUND: Topical and systemic drugs have been successfully used in the treatment of acne. However, many people are concerned about the side effects of these medicines, especially the childbearing women. Recent reports demonstrated that sequential treatment with laser- and light-based devices lead to a clinical improvement in acne. Recently, we witnessed experienced a case of improvement of inflammatory acne lesions during the treatment of acne scars using a 1,550-nm non-ablative fractional erbium-glass laser. OBJECTIVE: This study was designed to investigate the efficacy and safety of 1,550-nm non-ablative fractional erbium-glass laser in the treatment of facial inflammatory acne vulgaris. METHODS: 11 patients with facial inflammatory acne vulgaris were recruited. These patients received three treatment sessions at a 3-week interval. Inflammatory lesions were counted before and after treatment. The sebum production was quantified using the Sebumeter(R). We graded the patients' self-assessment and the investigator's global assessment using a five-point scale used by the dermatologist. We also investigated the histological changes after the treatment sessions, and the adverse effects during the study. RESULTS: Treatment with 1,550-nm non-ablative fractional erbium-glass laser was well tolerated, resulting in the reduction of inflammatory lesions by 61% (p<0.05). However, the reduction in sebum production from the baseline was not statistically significant. Histopathologic examination of the inflammatory lesions showed a marked decrease in the dermal inflammatory cell infiltration around the perivascular and periappendageal area and the sebaceous glands became smaller after laser treatments. Side-effects were minimal, and were resolved within a few days. CONCLUSION: 1,550-nm non-ablative fractional erbium-glass laser was safe and effective for the treatment of facial inflammatory acne lesions.

ABSTRACT

Acne vulgaris is one of the most frequent dermatoses in the general population. Numerous scientific articles are available on acne, mostly relating to its etiopathogeny. This notwithstanding, the large amount of scientific information generated by works on acne vulgaris has made it difficult to converge knowledge on its etiopathogeny into a single understanding. Therefore, this review has been proposed to analyze the four classic mechanisms of this dermatosis (sebum production, follicular hyperkeratinization, bacterial colonization and glandular inflammation), as well as its secondary mechanism, namely hormonal mediation.
